Role of Metabolism and DNA Adduct Formation in the Induction of Sister Chromatid Exchanges in Human Lymphocytes by Diethylstilbestrol1

چکیده
Recent studies have shown that diethylstilbestrol (DES) induces sister chromatid exchanges (SCEs) in lymphocytes from pregnant and premenopausal women but has only a slight effect on lymphocytes from postmenopausal women or men. In this study blood specimens from premenopausal women were used to define the role of different metabolic pathways on DES-induced formation of SCEs and to determine whether conditions resulting in induction of SCEs also induced detectable levels of DNA adducts. Exposure of lymphocytes in vitro to 0-40 /IM DES induced a concentration-dependent increase in SCEs. Addition of indomethacin to the cultures partially abolished DES-induced SCEs, sug gesting involvement of prostaglandin synthetases in the formation of specific DES metabolites that cause SCEs. a-Naphthoflavone, an inhib itor of cytochrome P-450 monooxygenases, had no effect on DES-induced SCEs. Cells exposed to DES at doses sufficient to cause large increases in SCE induction did not have adducts detectable by a "P-postlabeling assay capable of revealing adducts at a level of 1 adduct/10' normal

G2 chromosomal radiosensitivity and background frequency of sister chromatid exchanges of peripheral blood lymphocytes of breast cancer patients
Background: Chromosomal alterations play an important role in carcinogenesis. Enhanced chromosomal radiosensitivity is shown for many cancer predisposition conditions including breast cancer. In this study chromosomal radiosensitivity and the frequency of background sister chromnatid exchanges (SCE) in lymphocytes of normal individuals and breast cancer patients was compared. Materials...

متن کاملRelationships among benzo(a)pyrene metabolism, benzo(a)pyrene-diol-epoxide:DNA adduct formation, and sister chromatid exchanges in human lymphocytes from smokers and nonsmokers.
In the present study, benzo(a)pyrene (BP) metabolism, DNA adduct formation, ethoxyresorufin-O-deethylase activity, and sister chromatid exchange induction by BP were compared in human lymphocytes prepared from whole blood of smokers and nonsmokers following an in vitro incubation with BP. There was an approximate 7- to 10-fold variation in all parameters measured. To determine the source of thi...
متن کاملStudy of the Effect of MMC on the Sister Chromatid Exchange in the Human Lymphocytes
Some environmental mutagenic agents cause genomic instability and increase susceptibility of DNA damage. One of them is mitomycin C which is connected to DNA as an alkylating factor and affects susceptible cells to reduction reactions. This drug is used in chemotherapy and treatment of tumors. Study of genomic instability in the presence of different concentrations of MMC can show susceptibilit...
متن کاملSister chromatid exchanges, indices of human chromosome damage and repair: detection by fluorescence and induction by mitomycin C.
Sister chromatid exchanges in chromosomes from human lymphocytes grown two replication cycles in medium containing 5-bromodeoxyuridine can be detected by fluorescence microscopy after staining with the bisbenzimidazole dye 33258 Hoechst. These exchanges are much more frequent than chromosome or chromatid breaks and appear to be partly but not entirely due to 5-bromodeoxyuridine incorporation. S...
